The acid-cleavable Fd-Lip significantly upregulated the uptake (p<0.05 vs cFd-Lip at pH 7.4, p<0.01 vs Fd-Lip). The FA-Lip showed significantly higher uptake than PEG-Lip. Moreover, dual modification of FA and dNP2 further enhanced the cellular uptake, showing synergistic effects.

The endocytosis of cFd-Lip (pH 6.8) was higher than that of dNP2-Lip on folate receptor-positive 4T1 cells. The uptake of cFd-Lip by bEnd.3 monolayer cells was 1.01-, 1.26-, 19.90-, and 23.48-fold higher than that of Fd-Lip, dNP2-Lip, FA-Lip and PEG-Lip, respectively.
