Neurotoxin
FUNCTION: Neurotoxin that selectively increases Nav1.8/SCN10A peak current (EC(50)=894 nM) and causes a large hyperpolarising shift in both the voltage-dependence of activation and steady-state fast inactivation. Acts by binding to the domain II S3-S4 extracellular loop of the channel. In vivo, causes spontaneous pain behaviors (that gradually subsides over 30 minutes) when intraplantarly injected into mice. The same injection in Nav1.8/SCN10A knockout mice shows a reduction in pain behaviors, but not abolition, suggesting that activity on other Nav channels likely contributes to the nociceptive activity. {ECO:0000269|PubMed:35095499}.
